Trout imaging using OPT microscopy

Our colleagues from zebrafish phenotyping platform recently attended MiFoBio microscopy meeting. Taking with them freshly clarified rainbow trout (4g) provided by IERP, they had the opportunity to image the whole specimen on an home made Optical Projection Tomography (OPT) microscope.

Developed by Gaby G. Martins (Advanced Imaging Facility @IGC - Oeiras, Portugal), this OPenT setup is particularly adapted for large samples imaged which low magnification lenses (<4x). The trout (5 cm long, 1 cm large) was illuminated with one channel fluorescence emission mode (blue [LED] incident light & green fluorescence detected by the camera). Data processing was performed by Gabriel for 3D viewing of the entire trout acquired in only 20 minutes!!!!!!! Next step for images of clarified large samples would be to combine OPT to light sheet ultramicroscope.
